Cryomethods for Ultracryotomy and Immunolabeling
      (Workshop/Training)

      The 10-day program of the course consists of a balanced mix of lectures and demonstrations (25%) and practical work. The lectures will cover a broad spectrum of related topics. It will start with a 2 day optional part for participants who have no experience in standard EM techniques. ; The actual cryo-workshop will be given for 16 students (maximum).

In the first two days cryofixation and subsequent embedding techniques like freeze substitution, freeze-drying or progressive lowering of temperature will be dealt with in theory and practice. Training in ultracryotomy and immunolabeling will be the major issue in the remainder of the course. Under guidance of the skilled staff of the Utrecht Cell Microcopy Center, the complete Tokuyasu technique, including the latest modifications introduced in Utrecht, will be practiced on days 5 till 10.

The nitty-gritty of the technique is located in the lab and that’s where most of time will be spend. Biological samples and immuno-reagents will be available, but students are encouraged to bring their own diamond knife, (maximal 2) specimens and antibodies for sectioning and labeling during one of the later days. The Utrecht staff may be consulted for fixation of such material. ;
